Successful screening requires consideration of numerous factors, including the size of the experimental matrix and the volume of liquid required. High-throughput machines are needed as well. The 3 mm shaking throw of the Multitron Pro and Multitron Cell incubation shakers offer the perfect conditions for these applications, delivering screening results tailored to microorganisms and cell cultures, respectively.

Advantages of a 3 mm throw:
* Nearly four thousand batches in parallel
* Optimum oxygen introduction for relatively high cell densities and product yields
* Reduced liquid volume, which lowers the cost of culture media
* Evaporation effects eliminated through the use of direct steam humidification combined with suitable sample collection and sealing systems for microtiter or deep-well plates
 

Screening in Deep-Well-Platten

The use of deep-well plates significantly increases the likelihood of obtaining optimum screening results. This is reflected in the up to 8-fold increase in product yield that was observed in a study in which a Multitron Pro (deep-well plate) with a 3 mm shaking throw was compared to an incubation shaker with a 25 mm shaking throw.

As a result, researchers were able to obtain sufficient quantities of the target molecule from just 1 mL of liquid (deep-well plate) with nearly four thousand parallel batches. The larger culture vessels used up to now, by contrast, could only accommodate a few hundred batches.

 

Clone Screening followed by Small Scale GMP Manufacturing

The time for clone development can be reduced to 12 weeks using a three-stacked Multitron Pro with 3 mm shaking throw. The process starts in silico, and progresses through screening and clone selection in 96-deepwell plates up to five liter shaker flasks.

With up to 7,680 data points (80 microtiter plates) per shaker it is possible to manipulate multiple factors while performing all experiments in triplicates. This allows for the generation of statistically valid data for design of experiments (DoE) while also considering quality by design (QbD) as stable clones are generated and optimized.
